The intermediate undergoes enzymatic hydrolysis in an alkaline medium in which the acetoxy group of the molecule at position C3 of the aminocephalosporanic acid is hydrolyzed to form another intermediate with a free hydroxymethyl group. The intermediate is reacted with chlorosulfonyl isocyanate to form the corresponding N-chlorosulfonyl urethane. The compound is hydrolyzed by water to give the urethane. Finally, the benzhydryl protection is removed using trifluoroacetic acid to produce Cefuroxime.
Cefuroxime, commercially known as Ceftin, is a white to off-white crystalline powder having the molecular formula C16H16N4O8S and a molecular weight of 424.4 g/mol. It has a melting point in the range of 218-225 °C. It is freely soluble in the form of its sodium salt. It has an estimated water solubility of 0.284 mg/mL. The value of its dissociation constant, pKa, is 2.96. It is a cephalosporin antibiotic used to treat various bacterial infections.
