In the next step, butyrylalanine is subjected to a Dakin-West reaction with ethoxy oxalyl chloride in THF, using pyridine as a base and 4-dimethylaminopyridine as a catalyst, resulting in the formation of a mixed acid anhydride. Upon refluxing in ethanol, the mixture of amidrazone and the prepared mixed acid anhydride produced a condensation product. The dehydration-cyclization reaction with POCl3 formed imidazolotriazinone, which is then chlorosulfonated to produce an intermediate which followed by sulfonamide formation yields the desired Vardenafil.

Vardenafil, also known as Levitra, is a colorless solid, having the molecular formula C23H32N6O4S and a molecular weight of 488.6 g/mol. Its melting point is 192°C. Its solubility in water is 0.11 mg/mL (HCl salt). It has an estimated water solubility of 0.325 mg/ml at 25 °C. Its log P value is 1.4. It is a Phosphodiesterase type-5 inhibitor that is used to treat erectile dysfunction by increasing the blood flow. It can also be used to prevent and treat pulmonary hypertension and premature ejaculation.
